Simple and rapid LC-method for determination of moxifloxacin in human plasma: A bioequivalence study in healthy volunteers

Asimple isocratic reversed-phase high performance liquid chromatographic method with ultraviolet detection was developed and validated for determination of moxifloxacin in human plasma using gatifloxacin as an internal standard. The developed method was validated after optimization of various chromatographic conditions and other experimental parameters. Samples were separated using InertsilC18 (250 x 4.6mm, 5 ìm) analytical column. The mobile phase,methanol/30mMKH2PO4 buffer (pH2.5) (40:60, v/v) operated at 35oC column oven temperature was pumped at a flowrate of 1.0mL/min. The column eluents weremonitored at 300 nm. The presentmethod demonstrated acceptable values for selectivity, linearity within the expected concentration range (0.1-5 µg/mL; r2 = 0.9999 formoxifloxacin. Themethodwas efficiently applied to support a bioequivalence study of 400mgmoxifloxacin tablets in 24 healthy subjects.
